Fiz uma GUI com o Tkinter em python 3. Gostaria de saber se é possível ao fechar a janela, a aplicação fosse para o System Tray do Windows? Existe alguma lib ou comando dentro do Tkinter para isso.

Existem várias alternativas. Você pode usar o pystray (É necessário instalar o PIL):
pip install pystray
Ou o SysTrayIcon.py
É necessário instalar o pywin32 e ter um arquivo de ícone com o nome 'icon.ico' no diretório.
